Tybee is correct it was St Johns, but he is correct with both answers. Actual two other lights in Florida meet the criteria. Pensacola and Canaveral.

According to Wikipedia... The 45th parallel north is often called the halfway point between the Equator and the North Pole, but the true halfway point is actually 16.2 kilometers (10.1 miles) north of the 45th parallel because the Earth is oblate, that is, it bulges at the equator and is flattened at the poles. and In Michigan, the Old Mission Peninsula in Grand Traverse Bay ends just shy of the 45th parallel. Many guidebooks and signs at the Mission Point Lighthouse describe it as being halfway between the equator and north pole. When the Grand Traverse Bay recedes below normal level, it is possible to walk out to the exact line.

Scotland -Burntisland East Breakwater Light According to Lighthouse Directory (they have a picture too) Burntisland East Breakwater (relocated to Water of Leith) 1876. Inactive. Approx. 8 m (26 ft) octagonal cylindrical sheet iron tower with lantern and gallery, painted white with black trim. Scott Denham 's photo is at right, and Google has a satellite view. Thanks to Bill Newman for researching the identity of this lighthouse: it was installed at the end of the east breakwater at Burntisland. We don't know when it was deactivated, but it was relocated to Leith around 1990. Located at Tower Place on the west side of the Water of Leith, a small estuary that curves through the downtown area of the city. Site open, tower closed.

Three more possibilities.... all from Lighthouse Directory North Foreland (2) 1691 (height increased in 1793). Station established 1636. Active; focal plane 57 m (187 ft); five white flashes every 20 s. 26 m (85 ft) octagonal cylindrical stone tower with lantern and gallery, painted white, attached to two 2-story keeper's houses. A closeup photo is available, Trabas has a good photo, Werning has a photo, Huelse has a historic postcard view, Marinas.com has aerial photos, and Google has a satellite view. This historic lighthouse marks the beginning of the approach to the Thames estuary and the port of London. The tower was privately built and privately maintained until it was purchased by Trinity House in 1832. It was the last manned lighthouse in Britain, finally automated in November 1998. Located at the northeastern corner of Kent, on North Foreland Road 2 km (1.2 mi) north of Broadstairs. Site open; tower was formerly open, but is apparently now closed. Operator: Trinity House. Lowestoft (High) (4) 1874. Active; focal plane 37 m (121 ft); white flash every 15 s. 16 m (52 ft) round brick tower with lantern and gallery, painted white, attached to 2-story keeper's house. Ian Davey's photo is at right, a good closeup is available, Marinas.com has aerial photos, and Google has a satellite view. The former low light was discontinued and demolished in 1923. The lighthouse marks Lowestoft Ness, the easternmost point of the British Isles. It overlooks Sparrow's Nest Park and the Lowestoft and East Suffolk Maritime Museum, which sometimes conducts walking tours that include a visit to the light station. Located on Yarmouth Road (A12), on a bluff about 1.5 km (1 mi) north of the Lowestoft harbor entrance. On-street parking available. Site open, tower closed. Operator: Trinity House. While Bermudans claim Gibbs Hill LH(1844) is the oldest cast iron lighthouse in the Americas, the oldest is actually Morant Point LH (1841) in Jamaica.

Good guesses, but both the New Cape Henry and the last cast iron at Long Island Head were built in 1881 whereas the oldest U.S. surviving cast iron was built in 1846 and deactvated in 1954. To say the least it is not a well known light.

Engineer I.W.P. Lewis visited during his examination of the coast's lights in 1842. Lewis reported that the tower was leaky and the walls were cracked with frost. He added the following criticism:
A new cast-iron lighthouse -- the first of its type in the United States -- was built in the following year That would be 1843 or 44, that's why I included the link about Long Island Head Light... another site says.. In the U.S., the relatively low cost of cast iron probably appealed to Stephen Pleasonton, the Fifth Auditor of the Treasury and the pinch penny supervisor of U.S. lighthouses. In any case, he authorized the building of a small cast iron lighthouse at Long Island Head, Boston Harbor, in 1844. Prefabricated by the nearby South Boston Iron Company, the tower was built in three sections 7 feet high. It was 12 feet in diameter at the base and only 6 feet in diameter at the top, so the lantern, perched atop the narrow tower, gave the lighthouse a "big-headed" appearance. This lighthouse was demolished in favor of a larger cast iron tower in 1881. A second cast iron tower, essentially identical to the Long Island Head tower, was built in 1846 on Lake Champlain at Juniper Island, Vermont. The Juniper Island Light remained in service until 1954 and survives today in private ownership. That would make Juniper Island Light the 2nd one constructed

Reading the entire article,it appears to me that the earlier cast iron light was replaced by a newer cast iron in 1881 and thus it only had a life of 38 years. If it wasnt replaced, then it would be the oldest as well as the earliest. The question was which was oldest not the earliest.

According to the lighthousefriends website, the following sentence is written about the light I was seeking and it is not Long Island Head. Of course, they could be wrong.

MG, Need clarification here. You say the first US lighthouse agency, yet you specify COLONIAL lighthouses. Do you mean the lighthouses which were built by the British colonial government and were added to that US agency when created? Or do you mean an agency which oversaw the colonial lighthouses on future US soil? Taking a shot at the former, as I believe there was no colonial agency, all of these lights were either privately owned or built by individual colonies. The US Lighthouse Establishment was created by Congress in 1789. I believe there were 12 lighthouses and 7 spar buoys that were transferred to this branch. 8 of these lights were Boston, Brant Point, Beavertail, Sandy Hook, Tybee Island, New London Harbor, Charleston Harbor, Cape Cod

Very good job by Roland & Paul. US Lighthouse Establishment, 1789 & 12 are correct.
colonial (lower case) lighthouses is a description given in many books to the lighthouses built prior to the formation of the US and which were taken over by the USLHE IN 1789. There, of course, was no colonial agency.
Cape Cod is not correct as it was not established until 1797.
I've misplaced my book with the complete correct information, but I believe the others are correct. Paul is correct in that Newburyport was built in 1788 and I believe it was one of the 12, although not actually "ceded" (slow on the paperwork I expect) to the USLHE until 1790 according to another source I have. Cape Ann is definitely correct as it dates to 1771.
The only one that was missed is Plymouth, MA or Gurnet which was established in 1768.
Inasmuch as Roland provided the most information, he should probably go next unless he wishes to defer to Paul.

I have been out foxed.I was thinking of the Hudson River although the Detroit river meets my criteria, although my map shows only 7 lights. As to the Potomac unless you include the Chesapeake Bay I am not sure it applies. The Loon and Laura win. Either one submit the next question. Art

On Lighthouse Directory I also found: Canadian- Bar Point Pier Hackett Reach Range Rear Ballard Reef (Livingstone Channel Light 77D) US- Detroit Waterworks Intake Crib I am not positive that all these are actually in the Detroit River. Of the above, Grosse Ile, Boblo and Pelee Passage are inactive so they wouldn't count toward "coverage". I find it hard to determine what body of water a lighthouse is located in, for the Great Lakes. I have a book I could check, but my husband packed away all my lighthouse books while I was in Oregon, and I haven't had a chance to bring them back downstairs.

My research finds that the Potomac does not qualify as only Ft. Washington light is still active on the river. Inactive lights are Jones Point and Piney Point on the river and Point Lookout on the Chesapeake Bay at the entrance to the Potomac.

After due consideration I find due to a small technicality the Detroit river did not meet my criteria. I said a US river and since half of the Detroit river is in Canada it is not totally a US river. Art
